RhoA、RhoC基因在结直肠癌组织中的表达及其意义

目的 检测RhoA和RhoC mRNA在结直肠癌组织中的表达,并探讨其表达在结直肠癌发生、发展和侵袭转移中的意义.方法 TaqMan探针法进行实时荧光定量聚合酶链反应,检测RhoA和RhoC mRNA在正常黏膜组织、癌旁组织及结直肠癌组织中的相对表达,探讨与临床病理学指标的关系.结果 38例结直肠癌组织中RhoA和RhoC mRNA平均相对表达水平为正常黏膜组织的4.516和3.832倍,差异有统计学意义(P<0.05).RhoA、RhoC mRNA的表达均与有无淋巴结转移、有无肝转移有关(P<0.05).FdaoA mRNA的表达与组织学类型有关,Rhoc mRNA的表达与肠壁浸润深度有关(P<0.05).结论 RhoA和RhoC mRNA的表达在结直肠腺癌组织中明显增高,RhoA和Rhoc与结直肠癌发生、发展和侵袭转移密切相关.

Expression and significance of RhoA and RhoC in colorectal carcinoma

Objective To investigate the mRNA expression of RhoA and RhoC in colorectal carcinoma and its clinical significance. Methods The quantity of RhoA and RhoC mRNA relative to GAPDH mRNA was detected by TaqMan probe based real time PCR in 38 cases of colorectal carcinoma, corresponding para-cancer and normal tissues. The relationship between the quantity of RhoA and RhoC mRNA and some clinicopathological parameters were further analyzed. Results In colorectal carcinoma, the average quantities of RhoA and RhoC mRNA relative to GAPDH mRNA were significantly greater than in nor-mal tissues ( average relative expression was 4.516 and 3. 832 respectively, P < 0.05 ). The higher expression levels of RhoA and RhoC mRNA were related to lymph node and liver metastasis. The higher expression of RhoA mRNA in colorectal carcinoma was related to pathology and RhoC mRNA to depth of invasion. Conclusion The expression of RhoA and RhoC mRNA is significantly increased in colorectal carcinoma, which may play an important role in tumorigenesis and development, invasion depth and metastasis of colorectal carcinoma.
