摘    要:目的通过观察大鼠肝纤维化形成过程中胰岛素样生长因子-I受体(IGF—IR)的动态变化,探讨IGF-IR与肝纤维化的关系。方法制备四氯化碳诱导的大鼠肝纤维化模型,共8周。测定各组门静脉压,对肝组织进行常规苏木精-伊红和马松三色染色,采用免疫组织化学方法测定肝组织IGF—IR的表达,通过图像分析系统进行定量分析,并将IGF-IR与肝纤维化程度、门静脉压进行相关性分析。结果与正常对照组比较,模型组随着造模时间的延长,肝纤维化程度加重,门静脉压逐渐增高,IGF-IR在肝组织中的表达也增加,IGF-IR的表达与肝纤维化程度及门静脉压呈正相关。结论在肝纤维化进程中,IGF-IR表达逐渐增加,提示其可能参与肝纤维化。

Abstract:Objective To investigate dynamic alteration of insulin-like growth factor-I receptor (IGF-IR) during experimental liver fibrosis and study the association between IGF- I R and liver fibrosis. Methods Hepatic fibrosis was induced by chronic carbon tetrachloride (CCl4) subcutaneous injection for 8 weeks. The portal pressure was measured and conventional histological examinations of liver tissue were performed after hematoxylin and eosin staining and Masson staining. Hepatic tissue expression of IGF-1 R was detected by immunohistochemistry, and then quantitated by picture-analyzed systems. The correlations among the IGF- I R, the degree of liver fibrosis and the portal pressure were analyzed. Results In model group, liver fibrosis degree increased and portal pressure gradually increased as the time was extended. The IGF- I R expression was obviously higher than that in normal group, and it was positively correlated with liver fibrosis degree and the portal pressure. Conclusion The expression of IGF- I R in liver tissue increases with the development of hepatic fibrosis, which suggests it might participate in the hepatic fibrosis.
