细胞周期蛋白D1在大肠癌组织中的表达及其意义

目的检测细胞周期蛋白D1蛋白在大肠癌组织中的表达情况,并探讨其与大肠癌临床病理参数的关系。方法应用Western blot和免疫组织化学EnVision法检测50例大肠癌患者与10例外伤患者肠黏膜组织中细胞周期蛋白D1的表达水平,采用t检验和χ2检验进行统计学分析。结果细胞周期蛋白D1在大肠癌组织中表达主要在细胞核,也有少量表达在细胞质,阳性表达率为80.0%,明显高与在正常口腔黏膜组织中的表达率10.0%,差异有统计学意义(P=0.000);且随着大肠癌组织分化程度的降低,细胞周期蛋白D1的表达显著增加(r=0.972,F=119.304,Pr=0.000);淋巴结转移的大肠癌组织细胞周期蛋白D1表达高于无转移组织,差异有统计学意义(χ2=7.762,P=0.005);细胞周期蛋白D1在不同分期的大肠癌组织中表达不同(χ2=16.328,P=0.003),且细胞周期蛋白D1的表达与大肠癌的分期有关(r=-0.400,F=11.077,Pr=0.002)。结论细胞周期蛋白D1的异常表达与大肠癌的发生、发展有密切的关系,可作为大肠癌治疗和预后的辅助性指标。

Expression and its significance of Cyclin D1 in colorectal cancer

Objective To investigate the expression and significance of Cyclin D1 in colorectal cancer（CRC）.Method Western blot and immunohistochemistry method,EnVision,were employed to test the manifesting levels of Cyclin D1 in pathological slices of 50 CRC cases and 10 normal cases,and the results were analyzed using t-test and χ2 test.Results In 50 CRC cases,Cyclin D1 is mainly manifested in karyon,and a little is manifested in cytoplasm.The positive manifesting rates of Cyclin D1 in the CRC samples was 80.0%,which was notably higher than the manifesting rates of 10.0% in normal mucosal tissues,and the difference was notable（P=0.000）.As the degree of differentiation in CRC decreased,Cyclin D1 expression was significantly increased（r=0.972,F=119.304,Pr=0.000）.The manifestation of Cyclin D1 was correlated with the different pathological grades,clinical phases and lymph node metastasis.Conclusion The abnormal manifestation of Cyclin D1 was closely related with the occurrence and development of CRC.Therefore,Cyclin D1 can be used as subsidiary index for CRC treatment and prognosis.
