摘    要:目的研究Toll样受体9(TLR9)在中耳胆脂瘤上皮细胞的表达,并分析与白细胞介素1α(IL-1α)表达的关系。方法采用免疫组织化学二步法检测23例慢性中耳炎合并获得性胆脂瘤手术标本与10例胆脂瘤患者正常外耳道皮肤中TLR9、IL-1α的表达。结果在23例慢性中耳炎合并获得性胆脂瘤手术标本与10例胆脂瘤患者正常外耳道皮肤中,TLR9的表达指数分别为0.34±0.14、0.17±0.07,两者比较差异有统计学意义(P<0.01);IL-1α的表达指数分别为0.53±0.12,0.20±0.08,两者比较差异有统计学意义(P<0.01);胆脂瘤上皮TLR9与IL-1α的表达呈显著正相关(r=0.730,P<0.01)。结论 TLR9与IL-1α表达密切相关,表明TLR9可能上调IL-1α的表达,TLR9的信号转导参与了中耳胆脂瘤的骨质破坏过程。

Abstract:Objective To investigate the expression of Toll-like receptor 9 (TLR9) in middle-ear cholesteatoma, and to analyze its correlation with IL-1α expression. Methods Immunohistochemical SP method was used to examine the expressions of TLR9 and IL-1α in 23 surgically resected specimens of chronic otitis media combined with ac- quired cholesteatoma and another 10 of normal external ear canal skin from the cholesteatoma patients. Results The expression indices of TLR9 and IL-1α were 0.34±0.14 and 0.53±0.12 in 23 specimens of chronic otitis media com- bined with acquired cholesteatoma vs 0.17 ±0.07 and 0.20 ±0.08 in 10 of normal external ear canal skin from the cholesteatoma patients (P0.01). There was a significant linear correlation between TLR9 and IL-1α expressions (r= 0.730, P0.01) in cholesteatoma epithelium. Conclusion The close correlation between TLR9 and IL-1α expressions suggested that TLR9 may contribute to up-regulated expression of IL-1α in vivo, and that TLR9 signal transduction may be playing a role in bone destruction by middle-ear cholesteatoma.
