运动心脏重塑与微损伤发生中的细胞凋亡现象

摘    要:本研究选用健康成年雄性SD大鼠 70只 ,随机分为四种不同负荷运动训练组 ,建立运动心脏实验动物模型 ,采用电镜技术与流式细胞技术观察和分析在运动性心脏重塑与微损伤过程中 ,是否存在细胞凋亡现象以及细胞凋亡的发生与运动负荷及心肌微损伤的关系。结果显示 ,运动性的心脏结构和功能的重塑与心肌微损伤发生中存在心肌细胞凋亡现象 ,其细胞凋亡程度与运动强度有关 ;力竭运动时有、无训练大鼠心肌细胞凋亡的参与作用和程度存有差异 ,提示系统训练在一定程度上可以保护心肌细胞免受严重损伤 ,而运动性心肌微损伤的发生是由细胞坏死和细胞凋亡共同作用机制参与形成

Abstract:male Sprague-Dawley rats were randomly divided into sedentary control group(C), medium-intensity training group (MT), high-intensity training group (HT), once exhaustive exercise group (CE) and once exhaustive exercise after medium-intensity training group (ME). The rats of MT and HT groups run on a treadmill at the intensities of 60-80%VO 2max and 80-90%VO 2max for 12weeks, respectively.Based on successfully dissociating adult rat's cardiomyocytes into single cell suspension, and used advanced Flow Cytometry and DNA Multicycle Software to evaluate the apoptosis ratio. The results showed that apoptosis ratio in MT group had no significant difference from that of C group, while there was a significant increase in HT group as compared with C and MT groups. The apoptosis ratio also significantly increased in CE group. The ultrapathological observation showed that there were some myocyte apoptosis characteristics in HT and CE group. The results suggested that there was cardiomyocyte apoptosis in the exercise-induced cardiac remodeling and micro-injury.
