复方脑康胶囊对D-半乳糖老化小鼠海马神经元凋亡级联反应的影响

摘    要:目的:观察复方脑康胶囊水提液对D-半乳糖老化小鼠海马神经元凋亡级联反应的影响.方法:将小鼠随机分为5组:正常对照组(E组),D-半乳糖模型组(D组),复方脑康胶囊高、中、低剂量组(A,B,C组).每日sc D-半乳糖60 mg.kg-1造模,给模型小鼠复方脑康胶囊水提液(0.65,0.35,0.15g?kg-1 )ig,连续3个月.用免疫组织化学染色方法和Western blotting检测海马神经元凋亡诱导因子(AIF),Bax,细胞色素C(Cyt C)及Bcl-2的表达.结果:Western blotting结果显示,模型组Cyt c,Bax,Bcl-2表达量较正常对照组减少,而药物组表达量与正常对照组相似.免疫组织化学染色结果显示,模型小鼠海马神经元AIF,Bax,Cyt C的阳性细胞数分别比正常对照小鼠增加,而Bcl-2则比正常小鼠减少.与模型组相比,复方脑康胶囊水提液高、中、低3种不同剂量模型小鼠海马神经元AIF阳性细胞数从(24.7±6.1)个降低为(8.9±1.82),(12.6±4.51),(13.2±5.71)个;Bax阳性细胞数从(47.8±9.7)个降低为(11.2±3.2),(29.4±8.2),(31.1±6.3)个,Cyt C阳性细胞数从(36.7±9.1)个降低为(12.1±3.2),(20.2±6.8),(25.2±7.1)个,Bcl-2阳性细胞数从(10.4±6.4)个增加为(59.5±7.4)个,(48.6±9.2)个,(32.3±7.0)个.结论:复方脑康胶囊对D-半乳糖老化小鼠的海马神经元凋亡级联反应具有一定的改善作用.

Abstract:Objective: To observe the effect of compound Naokang capsule liquid extract on apoptosis cascade reaction of hippocampus neurons in mouse aging model induced by D-galactose. Method: The mice were randomly divided into 5 groups: normal control group (group E), D-galactose model group (D group), high dose group (A group), middle dose group (B group), low dose group (C group). 60 mg·kg-1 of D-galactose was sc given to the mice to produce AD model mice daily. Different doses of compound Naokang capsule liquid extract were given to model mice. The expression of apoptosis inducing factor (AIF) of hippocampus neuron,bax, cytochrome C(Cyt C) and Bcl-2 was tested by immunohistochemical staining and Western blotting. Result: The result of Western blotting showed that expression in the model group decreased, while the treatment groups were similar to the control group. The immunohistochemical staining results showed that, compared with control group, the number of AIF positive cells was reduced from 24.7±6.1 to 8.9±1.8,12.6±4.5,13.2±5.7; the number of Bax positive cells was reduced from 47.8±9.7 to 11.2±3.2,29.4±8.2,31.1±6.3, the number of Cyt C positive cells was reduced from 36.7±9.1 to 12.1±3.2,20.2±6.8,25.2±7.1, and the number of Bcl-2 positive cells was increated from 10.4±6.4 to 59.5±7.4,48.6±9.2,32.3±7.0. Conclusion: The apoptosis cascade reaction of hippocampus neurons in mouse aging model induced by D-galactose is obvious, which can be partly improved by compound Naokang capsule liquid extract.
