摘    要:目的 初步探讨细胞毒T细胞(CTL)对同种抗原的识别机制.方法 用负载特定肽的T2细胞与HLA-A2阴性个体的外周血淋巴细胞(peripheral blood lymphocytes,PBL)共培养,激活同种反应性CTL,通过乳酸脱氢酶(LDH)释放法观察同种反应性CTL对不同载肽T2细胞的杀伤活性.结果 用负载特定抗原肽的T2细胞诱导所获得的同种反应性CTL对负载不同抗原肽的T2细胞及空载T2细胞具有同等杀伤效应.结论 同种反应性CTL对同种MHC分子的识别不依赖抗原肽.

Abstract:Objective To investigate the mechanism of allogeneic recognition.Methods Peptides loaded T2 cells were used as stimulators to stimulate the HLA-A2~-donors peripheral blood lymphocytes(PBL) proliferating into alloreactive cytotoxic T lymphocytes(CTL).The alloreactive CTLs were used to test cytotoxicity with different targets(T2 cells with different peptides loaded).Results Alloreactive CTLs induced by the either 2 kinds of peptides loaded T2 had cytotoxicity targeting both peptides loaded T2 and null T2 with similar magnitude.Conclusion Allogeneic recognition of alloreactive CTLs to MHC molecules was antigenic peptide-independent.
