萎缩性胃炎组织病理背景胃癌的差异蛋白组学研究

摘    要:
目的通过新兴的蛋白组学方法分析筛选萎缩性胃炎向胃癌转变过程中关键的蛋白分子,探讨萎缩性胃炎癌变的分子机制。方法外科手术获取内镜及病理检查提示在萎缩性胃炎组织病理基础上发生的胃癌组织及癌周萎缩性胃炎组织。采用蛋白组学通用的双向电泳及质谱分析技术,筛选并鉴定胃癌组织及癌周萎缩性胃炎组织中差异表达的蛋白质。结果胃癌组织与癌旁萎缩性胃炎组织比较,共筛选出显著差异表达蛋白点29个,其中表达上调蛋白质点18个,表达下调蛋白点11个,这其中共有23个蛋白点被鉴定,上调蛋白14个,下调蛋白9个。结论在萎缩性胃炎基础上发生胃癌与其炎症组织背景的蛋白质表达存在显著差异,这些差异表达蛋白分子可能揭示萎缩性胃炎癌变的分子机制。

Abstract:
Objective To investigate the molecular mechanism underlying the transformation from chronic atrophic gastritis (CAG) to gastric cancer by a differential proteomic study.Methods A combination of two-dimensional gel electrophoresis and mass spectrometry was conducted to analyze the differentially expressed proteins between gastric cancer lesions and their CAG background.Results Twenty-three differentially expressed proteins were identified in gastric cancer compared with matched CAG mucosa,14 proteins were upregulated and 9 proteins were downregulated in gastric cancer compared with individual-matched CAG mucosa.Conclusion Proteins expression between gastric cancer and their CAG mucosa background are significantly different and the differentially expressed proteins may play some important roles in cancerization from CAG as functional molecules.
