牛磺酸对镧致仔鼠脑氧化损伤的改善作用研究

摘    要:目的 观察牛磺酸对镧致仔鼠脑氧化损伤的改善作用。方法 Wistar 孕鼠 40 只,建立仔鼠 0.5%LaCl3 水溶液镧暴露模型,于仔鼠生后49d随机分为阴性对照组、阳性对照组、低、中、高牛磺酸干预组,每组8只。阴性、阳性对照组用1.0ml/d蒸馏水灌胃,低、中、高牛磺酸干预组分别用200mg、400mg、800mg牛磺酸/kg·d灌胃,共计4w。采用Morris水迷宫评价仔鼠学习记忆能力,生化分析法测量仔鼠大脑皮质、海马谷胱甘肽(GSH)含量及过氧化氢酶(CAT)活力的表达水平。结果 Morris水迷宫中,染镧组仔鼠进入平台次数减少,出现学习记忆损伤,经牛磺酸干预后,仔鼠穿越隐蔽平台次数增加,差异有统计学意义(P<0.001)。染镧组仔鼠大脑皮质和海马的GSH 含量、CAT 活力均降低,差异有统计学意义(P<0.05)。干预后,各牛磺酸干预组仔鼠海马GSH 含量、CAT 活力均高于阳性对照组,差异均有统计学意义(P<0.05)。结论 牛磺酸能增加脑部抗氧化能力,对染镧仔鼠脑组织的氧化损伤具有改善作用,进而提高仔鼠学习记忆能力。

Abstract:Objective To observe the improved effect of taurine on brain oxidative damage induced by lanthanum.Methods Forty pregnant Wistar rats were used to establish lanthanum exposure model of 0.5%LaCl3 aqueous solution in their offspring.The rats were randomly divided into negative control group,positive control group(lanthanum-exposed group)and low,medium and high taurine intervention group at the end of 49 days after birth,8 animals per group.The negative and positive control group were treated with 1.0ml/d distilled water for 4 weeks.The low,middle and high taurine groups were treated with 200 mg,400 mg,800 mg of taurine/kg · d for 4 weeks respectively.The ability of learning and memory was evaluated by Morris water maze.The content of glutathione(GSH)and the expression of catalase(CAT)activity in cerebral cortex and hippocampus were measured by biochemical analysis.Results In the Morris water maze,the number of entering the platform decreased.Learning and memory impairment occurred in the lanthanum exposed group.After the intervention of taurine,the number of crossing the platform increased,the difference was statistically significant(P<0.001).The content of GSH and the activity of CAT in the cerebral cortex and hippocampus of rats exposed to lanthanum were decreased,the differences were statistically significant(P<0.05).After intervention,the content of GSH and the activity of CAT in hippocampus of each taurine intervention group were higher than those of the positive control group,the differences were statistically significant(P<0.05).Conclusion Taurine can increase the antioxidant capacity of the brain and improve the oxidative damage of the brain tissue of the lanthanum-exposed rats and the learning and memory ability of the rats.
