褪黑素对梗阻性黄疸大鼠小肠黏膜屏障保护作用的实验研究

摘    要:目的探讨褪黑素对大鼠梗阻性黄疸后小肠黏膜屏障的保护作用。方法将36只大鼠随机分为3组(每组12只):假手术对照(SO)组、梗阻性黄疸(OJ)组、褪黑素治疗(MT)组。观察各组血浆内毒素、肿瘤坏死因子(TNF-α)、白细胞介素6(IL-6)、小肠组织中细胞间黏附分子(ICAM-1)的表达及小肠组织形态学改变,并测量肠绒毛高度和黏膜厚度。结果OJ组血浆内毒素、TNF-α、IL-6明显增高,小肠组织ICAM-1的表达明显增强,肠绒毛高度、黏膜厚度明显降低,与SO组有显著性差异(P<0.01);MT组与OJ组比较血浆内毒素、TNF-α、IL-6降低(P<0.01),小肠组织ICAM-1的表达明显减弱(P<0.01),肠绒毛高度、黏膜厚度增高,肠组织结构损害减轻(P<0.01)。结论褪黑素通过减少肠道细菌内毒素的移位,降低血浆中细胞因子水平,抑制梗阻性黄疸后肠组织ICAM-1的表达,保护小肠黏膜屏障。

Abstract:Objective It is to discuss the protective action of melatonin on small intestine mucosa barrier of obstructive jaundice rats.Methods 36 rats were randomly divided into sham operated group(SO),obstructive jaundice group(OJ) and melatonin treatment group(MT).The levels of plasma endotoxin,TNF-α and IL-6,ICAM-1 expression and small intestine tissue morphological change were observed.The intestinal villus height and mucosal thickness were measured.Results Compared with SO group,the levels of plasma endotoxin,TNF-α and IL-6 in OJ group obviously heightened(P<0.01),ICAM-1 expression in OJ group obviously strengthened(P<0.01) and the intestinal villus height and mucosal thickness in OJ group obviously lowered(P<0.01).Compared with OJ group,the levels of plasma endotoxin,TNF-α and IL-6 in MT group lowered(P<0.01),ICAM-1 expression in MT group obviously weakened(P<0.01),the intestinal villus height and mucosal thickness in MT group heightened(P<0.01) and the damage of intestinal tissue structure lightened(P<0.01).Conclusion Melatonin can protect small intestinal mucosa barrier through decreasing the shift of intestinal bacteria endotoxin,lowering cell factor level in plasma and inhibiting ICAM-1 expression in intestinal tissue after obstructive jaundice.
