水蛭提取液体外对人视网膜色素上皮细胞PAR-1表达的影响

摘    要:目的:研究水蛭提取液对人视网膜色素上皮(retinal pigment epichelial,RPE)细胞PAR-1表达的影响。方法:水蛭提取液体外单独或与凝血酶共同作用于人RPE细胞后,用免疫荧光法测定PAR-1的荧光表达。结果:PAR-1的免疫荧光光密度值比较:(1)凝血酶组与正常对照组比较,PAR-1的IOD值下降(1608±675,4036±1134,P<0.01)。(2)单加水蛭提取液组与正常对照组比较,PAR-1的IOD值升高(9998±2374,4036±1134,P<0.01)。(3)凝血酶和水蛭提取液同时加药组与凝血酶组比较,PAR-1的IOD值显著升高(19664±5436,1608±675,P<0.01)。表明水蛭提取液能竞争性抑制凝血酶对PAR-1的活化作用。结论:水蛭提取液能抑制凝血酶诱导的人RPE细胞膜上的PAR-1的活化,阻断PAR-1介导的细胞信号传导。

Abstract:AIM:To research the influences of leech extracting solution on expression of thrombin receptor-1 in cultured human retinal pigment epithelial(RPE) cells.·METHODS:After RPE cells were treated by leech extracting solution alone or leech extracting solution with thrombin,flourescence expression of PAR-1 was mea-sured by immumofluorescence method.·RESULTS:Flourescence integral optical density(IOD) of PAR-1 displayed that thrombin group: compared with normal control group,IOD decreased (P<0.01);leech extract group: compared with normal control group,IOD increased(P<0.01);leech extracting solution with thrombin group:compared with thrombin group,IOD significantly increased (P<0.01). ·CONCLUSION:Leech extracting solution seems to be a competitive inhibitor of thrombin during PAR-1 activation in hRPE cells.It can inhibit the activation of PAR-1 induced by thrombin in human RPE cell membrane and interrupt PAR-1-mediated cell signal transduction.
