离体大鼠心脏胰岛素抵抗与心肌损伤

摘    要:
目的探讨高胰岛素刺激后离体大鼠心脏发生心肌胰岛素抵抗与心肌损伤。方法30只SD大鼠按灌注液中胰岛素含量随机均分为0U/L组(Ⅰ组)、10U/L组(Ⅱ组)、20U/L组(Ⅲ组)、30U/L组(Ⅳ组)和50U/L组(Ⅴ组)五组。采用Langendorff模型以含不同浓度胰岛素的K-H液进行离体心脏灌注。定时搜集并测定冠状动脉流出液样本中葡萄糖浓度,评估心肌葡萄糖氧化情况;测定流出液中肌酸激酶(CK),肌钙蛋白I(cTnI)含量;Western Blot检测心肌细胞胰岛素受体(InsR)含量。结果与Ⅰ组比较,Ⅱ~Ⅴ组大鼠心肌葡萄糖总氧化量明显减少(P<0.05),心肌细胞InsR表达降低,心肌细胞释放CK和cTnI增加(P<0.05)。结论高胰岛素刺激大鼠离体心脏后,心肌细胞出现胰岛素抵抗现象。其程度随胰岛素含量升高而增强,心肌亦受到损伤。

Objective To confirm a hypothesis of myocardial insulin resistance phenomenon on isolated rat heart after stimulated with hyperinsulinism.Methods Thirty SD rats were randomized into five groups according to the concentration of insulin in the buffer.
