休止期毛乳头细胞差异表达基因的分析

摘    要:
目的筛选休止期毛乳头细胞差异表达基因,探讨脱发机制.方法从同一斑秃患者头皮获得脱发区和非脱发区毛乳头,应用SMART cDNA和抑制性消减杂交建立消减文库,对部分克隆进行杂交筛选、测序及同源性检索分析.结果成功构建了休止期(斑秃脱发区)毛乳头cDNA消减文库,代表毛乳头在休止期表达上调的基因,测序发现1条与甲状腺相关的自身抗原基因,还发现脱发区毛乳头差异表达与抑制性信号转导途径有关的基因.这些基因多是首次发现在毛囊中有表达.结论斑秃毛乳头可能由于表达自身抗原基因而导致针对其的自身免疫反应的启动,后者通过多种信号转导途径影响毛乳头的生长和功能发挥.

Abstract:
Objective To screen and clone differentially expressed genes in dermal papilla from follicles at telogen and to explore the mechanism of alopecia. Methods Dermal papillae of lesional and normal follicles were separated from the scalps of alopecia areata that occurred in a twenty-eight male. The cDNA fragments of differentially expressed genes in dermal papilla from lesional and normal follicles of alopecia areata were isolated by suppression subtractive hybridization. Positive clones were screened by PCR method and verified by cDNA dot blotting and then analyzed through homologous retrieving. Results An efficient subtractive library of dermal papillas of lesional follicles from alopecia areata, which contained differentially genes in dermal papillae at telogen, was established. It was showed that dermal papillas at telogen could express an autoantigen gene and others genes related to inhibiting signal transduction. Conclusion The autoimmune response directed to dermal papillas, which would inhibit the growth and function of dermal papillas, might be induced due to the expression of autoantigen at catagen.
