瞬时受体电位通道蛋白6(TRPC6)在SGC-7901细胞增殖中的作用

目的 探讨瞬时受体电位通道蛋白6(TRPC6)与SGC-7901细胞增殖的关系。方法 体外培养SGC-7901细胞,以是否加入TGF-β₁分为对照组和实验组。根据实验组是否加入SKF96365抑制剂分为TGF-β₁和TGF-β₁+SKF96365组。免疫荧光染色观察TRPC6在对照和TGF-β₁组的表达。Western blot方法检测TRPC6和增殖指标在对照组、TGF-β₁组和TGF-β₁+SKF96365组中的变化。结果 与对照组相比,TRPC6蛋白在TGF-β₁组中表达升高(P＜0.05)。抑制TRPC6蛋白表达,增殖指标在TGF-β₁+SKF96365组表达明显降低(P＜0.01)。结论 抑制TRPC6蛋白表达可降低SGC-7901细胞增殖能力。

Effect of transient potential receptor channel protein 6 (TRPC6) on proliferation of SGC-7901 cells

Objective To investigate the expression of TRPC6 in SGC-7901 cells and to elucidate the relationship between TRPC6 protein and proliferation of SGC-7901 cells.Methods SGC-7901 cells were cultured in vitro,and SGC-7901 cells were divided into control group and experimental group with or without TGF-β₁ treatment. The experimental groups were divided into TGF-β₁ and TGF-β₁+SKF-96365 group according to whether incubated with SKF96365 or not.Immunofluorescence staining was used to observe TRPC6 protein expression in SGC-7901 cells in the control and TGF-β₁ groups.Western blot was used to detect the protein changes of TRPC6 and proliferation indexes in control group,TGF-β₁ group and TGF-β₁+SKF96365group.Results Compared with the control group,the expression of TRPC6 protein in TGF-β1 group was significantly increased(P＜0.05).SKF96365 inhibited TRPC6 protein expression,significantly suppressed the proliferation of SGC-7901 cells(P＜0.01).Conclusion Blockade of TRPC6 protein expression could reduce the proliferation ability of SGC-7901 cells.
