前列腺素E1对梗阻性黄疸大鼠肠黏膜损伤的保护作用

摘    要:目的研究前列腺素E1对梗阻性黄疸大鼠肠黏膜损伤的保护作用。方法将45只Wistar雄性大鼠按随机数表法分为假手术组(Sham组)、梗阻性黄疸组(OJ组)、前列腺E1组(OJ+PGE1组),每组15只,采用胆总管双重结扎建立大鼠梗阻性黄疸模型,术后第7天起,OJ+PGE1组应用前列腺素E1注射液0.1μg·kg-1·d-1腹腔注射治疗14 d,其他两组腹腔注射相同剂量生理盐水。3组大鼠分别于术后第21天处死,利用酶联免疫吸附法(ELISA)检测和比较3组大鼠血清内毒素、D-乳酸、TNF-α、IL-6的水平,HE染色观察比较各组大鼠回肠绒毛高度、肠黏膜厚度,免疫组织化学染色和Western-blot检测回肠组织NF-κB蛋白的表达。结果 OJ+PGE1组及OJ组大鼠血浆内毒素、D-乳酸、TNF-α、IL-6的水平表达水平高于Sham组,但OJ+PGE1组增高的程度低于OJ组,差异有统计学意义(P<0.05);与Sham组比较,OJ+PGE1组与OJ组大鼠回肠组织NF-κB蛋白表达明显升高,但OJ+PGE1组增高的程度低于OJ组,差异有统计学意义(P<0.05)。结论前列腺素E1对梗阻性黄疸大鼠肠黏膜损伤具有保护作用,其机制可能与抑制NF-κB的活化和降低炎性因子的表达有关。

Abstract:Objective To study the protective effect of prostaglandin E1 on intestinal mucosal lesion in rats with obstructive jaundice. Methods A total of 45 male Wister rats were randomly divided into the sham operation group (SH group), obstructive jaundice group (OJ group) and obstructive jaundice + prostaglandin E1 treatment group (OJ+PGE1 group), with 15 rats in each group. The SH group rats were free without ligation of the common bile duct, OJ group and Group OJ+PGE1 were treated by double thread ligation of the common bile duct, postoperative 1 week, OJ+PGE1 group application of prostaglandin E1 injection (0.1 μg·kg-1·d-1) continuous intraperitoneal injection for 2 weeks. The other two groups were intraperitoneally injected with the same dose of normal saline. The three groups of rats were sacrificed on the twenty-first day, expression of serum endotoxin, D-lactic acid, TNF-α, IL-6 were measured by enzyme-linked immunosorbent assay (ELISA), the ileum villus height and thickness of the rats were compared by HE, the expression of nuclear factor-κB (NF-κB) protein were detected by immunohistochemical staining and Western-blot. Results The expression level of serum endotoxin, D-lactic acid, TNF-alpha and IL-6 in the OJ+PGE1 group and OJ group were higher than the Sham group, but the OJ+PGE1 group was significantly lower than the OJ group (P<0.05); compared with the Sham group, the expression of NF-κB protein in the OJ+PGE1 group and OJ group were increased significantly, but the increase in the OJ+PGE1 group was significantly lower than the OJ group (P<0.05). Conclusion Prostaglandin E1 has protective effect on intestinal mucosal injury in rats with obstructive jaundice, and its mechanism may be related to inhibiting the activation of NF-κB and decreasing the expression of inflammatory factors.
