含鼠白细胞介素12基因腺病毒载体对小鼠支气管哮喘模型肺组织GATA-3表达的影响

摘    要:目的:探讨含鼠白细胞介素12(IL-12)基因的腺病毒载体(AdCMVIL-12)对小鼠支气管哮喘模型GATA-3表达的影响。方法:BALB/c小鼠30只,随机分为正常对照组(A组)、哮喘模型组(B组)和AdCMVIL-12实验组(C组),B和C组建立哮喘模型,并于第20天分别鼻内吸入50 μL生理盐水和5×108 PFU AdCMVIL-12。第28天收集各组小鼠的支气管肺泡灌洗液(BALF),比较细胞总数及细胞分类的变化,HE染色观察小鼠支气管及肺组织的病理变化,免疫组化及RT-PCR方法检测各实验组肺组织中GATA-3的表达情况。结果:哮喘模型组与对照组及AdCMVIL-12实验组比较,小鼠肺组织有大量炎症细胞浸润,以嗜酸性粒细胞为主。免疫组织化学及RT-PCR结果显示,哮喘模型组小鼠肺组织GATA-3表达(0.48 ±0.06)明显高于实验组(0.16±0.07)及对照组(0.18±0.04 )(P<0.01)。结论:AdCMVIL-12对小鼠支气管哮喘的气道及肺组织炎症有明显的抑制作用,其机制可能与AdCMVIL-12阻断GATA-3的表达,调节Th1/Th2细胞因子的平衡有关。

Abstract:Objective To investigate the effects of adenovirus vector carrying murine interleukin-12 gene(AdCMVIL-12) on the expression of GATA-3 in mouse model with asthma.Methods Thirty BALB/c mice were randomly divided into 3 groups: control group(group A),asthma model group(group B) and AdCMVIL-12 experimental group(group C).Asthma models were established in group B and C,saline(50 μL) and 5×108 PFU AdCMVIL-12 were given by intranasal administration at the 20th days.BALF were recovered at the 28th day for cellular classification.Pathological changes of bronchial and lung tissue were observed by HE staining in each groups.The expression of GATA-3 in each group was detected by immunohistochemistry and RTPCR.Results Compared with group A and C,a lot of inflammatory cells were found in group B and the percent of eosinophils was the most,the expression of GATA3 in lung tissue of group B(0.48 ±0.06) was higher than those in group C(0.16±0.07) and group A(0.18±0.04)(P<0.01).Conclusion AdCMVIL-12 can significantly inhibit airway and lung tissue inflammation in asthma model,its mechanism may be related to blocking the expression of GATA-3 in asthma model and regulating the balance of Th1/Th2 cytokines.
