Cortactin、N-WASP及PTTG1IP蛋白在不同类型结肠组织中的表达及意义

摘    要:
目的 探讨cortactin、N-WASP和PTTG1IP蛋白在不同类型结肠组织中的表达及意义。方法 应用免疫组织化学方法检测25例正常结肠、25例结肠腺瘤和50例结肠癌组织中cortactin、N-WASP和PTTG1IP蛋白表达情况。利用卡方检验分析cortactin、N-WASP和PTTG1IP蛋白在不同结肠组织中的表达差异及与结肠癌患者临床病理特征间的关系。利用Spearman秩相关检验分析cortactin与N-WASP及PTTG1IP表达的相关性。结果 正常结肠组织、结肠腺瘤和结肠癌组织中,cortactin蛋白表达率分别为44.00%、40.00%、72.00%;N-WASP蛋白表达率分别为16.00%、4.00%、44.00%;PTTG1IP蛋白表达率分别为32.00%、64.00%、74.00%。Cortactin及N-WASP蛋白表达率在结肠癌组织均显著高于正常结肠和结肠腺瘤组织(P<0.05);PTTG1IP蛋白表达率在结肠癌和结肠腺瘤组织均显著高于正常结肠组织(P<0.05)。结肠癌组织中,cortactin及PTTG1IP蛋白表达均与淋巴结转移及TNM分期有关(P<0.05),且两者表达呈正相关(r=0.341,P<0.05)。结论 Cortactin、N-WASP及PTTG1IP蛋白在结肠癌的发生中均发挥作用。Cortactin和PTTG1IP蛋白在结肠癌的进展中可能发挥协同促进作用,并可能成为治疗结肠癌的新靶点。

Abstract:
Objective To detect the expression and significance of cortactin, N-WASP and PTTG1IP proteins in different types of colon tissue. Methods Immunohistochemical staining was used to detect the protein expressions of cortactin, N-WASP, PTTG1IP in 25 specimens of normal colon, 25 specimens of colon adenoma and 50 specimens of colon cancer. Chi-square test was used to analyze the differential expression of cortactin, N-WASP and PTTG1IP in different groups, and the relationships between cortactin, N-WASP, PTTG1IP expressions and clinicopathological features of colon cancer patients. Spearman rank correlation test was used to analyze expression correlations of cortactin with N-WASP and PTTG1IP. Results In normal colon, colon adenoma and colon cancer tissues, the expression rates of cortactin protein were 44.00%, 40.00%, and 72.00%; N-WASP 16.00%, 4.00%, and 44.00%; PTTG1IP 32.00%, 64.00%, and 74.00%, respectively. The expression rates of cortactin and N-WASP were significantly higher in colon cancer group than those in normal colon and colon adenoma groups (P<0.05). The expression rates of PTTG1IP in colon cancer and colon adenoma groups were significantly higher than that in normal colon group (P<0.05). In colon cancer group, the protein expressions of cortactin and PTTG1IP were significantly correlated (P<0.05) and both associated with lymph node metastasis and TNM stage (P<0.05). Conclusions Cortactin, N-WASP and PTTG1IP proteins may play roles in colon cancer tumorigenesis. Cortactin and PTTG1IP proteins may play synergistical effect on promoting the progression of colon cancer and may represent new therapeutic targets for colon cancer.
