表皮生长因子(EGF)在人睾丸中的表达

摘    要:为了解人睾丸是否产生EGF及其细胞定位,本实验应用鼠抗人EGF单克隆抗体免疫组织化学方法,研究了人胚、正常成人、隐睾和精原细胞癌患者睾丸的EGF表达和变化。免疫组化分析的结果表明:(1)26周人胚睾丸间质细胞呈EGF弱阳性反应。(2)正常成年男性睾丸的多数间质细胞是EGF强阳性染色,部分精原细胞和个别曲细精管周边的肌样细胞是EGF阳性染色。睾丸支持细胞未见明显阳性反应。(3)隐睾组织整体水平上的EGF阳性间质细胞数量减少,尽管在个别区域出现聚积的间质细胞团,其中部分间质细胞呈EGF强阳性染色,部分间质细胞反应较弱。(4)恶性增生的精原细胞是EGF强阳性表达。上述结果证明:人睾丸是可以产生EGF的,间质细胞是其主要的EGF分泌细胞。隐睾组织中表达EGF的间质细胞减少,提示隐睾症的生精障碍可能与EGF的表达减少有关。

Abstract:The human testes from fetus,adults and the subjects suffering from cryptorchidism or spermatocytoma were studied immunohistochemically to determine whether human testis can serve as a source of epidermal growth factor (EGF), what kind of cells can produce EGF,and how EGF expression is changed in some pathologic states. The results demonstrated that (1)EGF immunostaining showed weak positivity on Leydig cells in human testis from the 26 -week - old fetus; (2) in human testis from adults strongly intense positivity appeared in the majority of Leydig cells, a part of spermatogonia and occasionally in a few peritubular cells while Sertoli cells did not show any positive staining; (3) in the undescending testis, although the accumulated Leydig cell group appeared occasionally which was composed of some strongly immunoreactive cells and some weakly stained cells, the number of positively stained Leydig cells decreased as a whole; and (4)strong immunstaining was observed in the cytoplasm of the spermatogonia from malignant spermatoctoma; This study revealed that human testis can produce EGF mainly from Leydig cells,and a decrease in the number of Leydig cells expressing EGF in the undescending testis indicated that infertility of cryptorchid may be related to the reduction of EGF secreted by testis itself.
