人参皂甙Rh2诱导人喉癌细胞株Hep-2凋亡的研究

目的:研究人参皂甙Rh-2(G-Rh2)诱导喉癌Hep-2细胞凋亡的作用。方法:应用流式细胞术、琼脂糖凝胶电泳检测G-Rh2处理前后Hep-2细胞的凋亡情况,并应用SP免疫组化法检测药物处理前后相关基因产物Bax、Bcl-2蛋白的表达,观察G-Rh2对喉癌Hep-2细胞的诱导凋亡作用及其对细胞周期的影响。结果:(1)G-Rh2具有诱导凋亡作用,且呈量效、时效关系,细胞凋亡率随G-RH2浓度及作用时间的增加而升高,并可将细胞周期阻滞于G0/G1期。(2)G-Rh2处理Hep-2后,Bax表达上调,Bcl-2在G-Rh2处理前后无明显变化。结论:G-Rh2对体外培养的Hep-2细胞具有诱导凋亡的作用,其机制可能与上调Bax的表达及细胞周期阻滞有关。

STUDY ON THE EFFECTS OF GINSENISIDE-RH2 ON INDUCING HUMAN LARYNGEAL CARCINOMA CELL LINE HEP-2 APOPTOSIS

Objective:To study the effects of ginsenoside-Rh2(G-Rh2) on inducing human laryngeal carcinoma cell line Hep-2 apoptosis.Methods: Flow cytometry and agarose gel electrophoresis were used to detect the apoptosis of Hep-2 before and after G-Rh2 was used.SP immunohistochemical technique was used to observe the expression of related gene products: Bax and Bcl-2.Results:(1) G-Rh2 induced apoptosis of Hep-2 cell line in a dose-and time-dependent manner,and the apoptotic rate of Hep-2 cells line increased with the increasing of density and action time of G-Rh2.The cell cycle was stopped at G_0/G_1 stage.(2) The expression of Bax was up-regulated after G-Rh2 was used,but the expression of Bcl-2 had no obvious changes.Conclusions: G-Rh2 could induce Hep-2 cell line apoptosis,and its mechanisms maybe related to up-regulating Bax expression and stop cell cycle.
