登革2型病毒及其NGC株、M株E基因的原核蛋白对HUVEC通透性的影响

摘    要:目的:研究登革2型病毒(DENV-2)、登革2型病毒国际参考株(NGC)E基因1-476序列原核蛋白、登革2型病毒贵州蚊虫分离株(M)E基因1476序列原核蛋白对人脐静脉内皮细胞(HUVEC)通透性的影响.方法:将DENV-2 M株和NGC株E基因1-476序列原核表达产物及DENV-2接种于HUVEC单层细胞,用tran-swell法检测HUVEC的通透性,透射电镜观察细胞超微结构的改变.结果:DENV-2、NGC株E基因的原核蛋白、M株E基因的原核蛋白分别在30 min、3h、12 h时对HUVEC通透性的影响最为明显,透射电镜结果显示有细胞超微结构的改变.结论:DENV-2、DENV-2 NGC株E基因原核蛋白、M株E基因原核蛋白对HUVEC通透性、细胞超微结构有明显影响.

Abstract:Objective: To study the effect on human umbilical vein endothelial cell (HUVEC) per- meability by Dengue Virus type 2 (DENV-2)and prokaryotic protein of 1 -476 sequences of dengue vi- rus type 2 M and NGC strains. Methods: HUVEC monolayer cells were vaccinated with E gene 1 - 476 sequence prokaryotic expression products of strains DENV-2 M and DENV-2 NGC, as well as DENV-2. The HUVEC permeability was tested by Transwell method, and transmission electron micro- scope (TEM) was used to observe ultrastructure changes of cells. Results: Effects on HUVEC perme- ability of DENV-2, NGC strain E gene prokaryotic protein and M strain E gene prokaryotic protein were most obvious at 30 minutes, 3 hours, and 12 hours of the experiment respectively. TEM results presented uhrastructure changes of cells. Conclusion: HUVEC and HUVEC permeability can be obvi- ously influenced by DENV-2, and E gene prokaryotic proteins of DENV-2 NGC strain and M strain.
