钩吻根脂溶性化学成分的分离与鉴定

摘    要:目的对钩吻根脂溶性部位的化学成分进行研究。方法通过乙醇回流提取、溶剂分步萃取得到钩吻根粗提物,利用硅胶柱、ODS柱、Sephadex LH-20、Toyopearl HW-40F等多种色谱柱进行分离纯化,利用NMR等现代波谱技术对分离得到的化合物进行结构鉴定。结果分离得到10个化合物,分别鉴定为钩吻素子(koumine,1)、钩吻绿碱(gelsevirine,2)、胡蔓藤碱乙(humantenine,3)、钩吻内酰胺(gelsemamide,4)、12β-羟基-孕甾-4,16-二烯-3,20-二酮(12β-hydroxy-pregna-4,16-diene-3,20-dione,5)、正三十一烷醇(n-hentriacontyl alcohol,6)、十四碳酸甲酯(methyl myristate,7)、6,7-二甲氧基香豆素(6,7-dimethoxycoumarin,8)、3,4,5-三甲氧基苯甲醇(3,4,5-trimethoxybenzyl alcohol,9)、5-甲基间苯二酚(5-methylresorcinol,10)。结论化合物6-10为首次从胡蔓藤属中分离得到。

Abstract:Objective To investigate the liposoluble constituents from the roots of Gelsemium elegans Benth.. Methods The aqueous ethanol extract from the roots of G. elegans was isolated and purified with silica gel, ODS, Sephadex LH-20 and Toyoperal HW-40F column chromatography. Physicochemical properties and spectroscopic analysis including 1D, 2D NMR and MS were employed for the determination of the structures. Results Ten compounds were isolated,and their structures were identified as koumine (1), gelsevirine (2), humantenine (3), gelsemamide (4), 12β-hydroxy-pregna-4,16-diene-3,20-dione (5), n-hentriacontyl alcohol (6), methyl myristate (7), 6,7-dimethoxycoumarin (8), 3,4,5-trimethoxybenzyl alcohol (9) and 5-methylresorcinol (10). Conclusions Compounds 6-10 are obtained from the genus Gelsemium for the first time.
