自体静脉套管联合雪旺细胞移植对周围神经缺损再生的影响

摘    要:
目的观察自体静脉套管联合雪旺细胞移植对周围神经缺损再生的影响,以探索周围神经缺损修复的新的治疗方法。方法20只新西兰白兔随机分为实验组和对照组,每组10只。取兔自体股外静脉,套于神经两断端之间,制成静脉套管,实验组向静脉套管内注射雪旺细胞、对照组注射生理盐水,术后6周通过光镜及透射电镜观察神经再生情况,采用SPSS 13.0软件包对数据进行统计分析,组间均值比较采用t检验,P〈0.05为差异有统计学意义。结果HE染色结果显示,实验组有髓神经和无髓神经数量较多,排列较整齐,髓鞘清晰可见,纤维数量较少。对照组有一定数量无髓神经,而有髓神经少见,纤维数量较多。t检验结果显示差异具有统计学意义(P〈0.05)。电镜观察实验组有髓神经纤维数量较多,排列较整齐,髓鞘清晰可见,纤维数量较少。可见雪旺细胞与有髓神经纤维相连。对照组有髓神经纤维和无髓神经纤维数量较少,排列不整齐,髓鞘不清晰,纤维数量较多,无雪旺细胞。结论本研究结果表明静脉套管可以促进周围神经再生;雪旺细胞注入神经缺损的静脉套管对周围神经再生的作用优于单纯使用静脉套管。

Abstract:
Objective To investigate the effect of Schwann cell transplantation on peripheral nerve regeneration.Methods Twenty New Zealand white rabbits were randomly divided into experimental group and control group.The autogenous femoral vein of rabbit was isolated,resected and then interposed between the nerve stumps.Schwann cells(experimental group) or normal saline(control group) were injected into the vein conduit of the rabbit.The nerve regeneration was observed under light and electron microscopes 6 weeks after the surgery.Software SPSS13.0 was used to process the data.P〈0.05 was considered as significant.Results More regularly arranged myelinated and unmyelinated nerve fibers were observed in experimental group than those in control group.Complete myelination and fewer fibers were found in the experimental group compared with those of control group.Schwann cells were found to connect with myelinated nerve fibers in the experimental group,while no Schwann cells were found in the control group.Conclusion Autogenous vein conduit improved the regeneration of peripheral nerve.The vein conduits seeded with Schwann cells has greater effect on peripheral nerve regeneration than the conduits alone.
