神经肽Y对抑郁症模型大鼠大脑皮质NE和5-HT表达的影响

摘    要:
目的探讨神经肽Y(NPY)对抑郁症模型大鼠大脑皮质去甲肾上腺素(NE)和5-羟色胺(5-HT)表达的影响。方法将30只SD大鼠随机分成3组:NPY干预组,生理盐水干预组和空白对照组;对NPY干预组和生理盐水干预组大鼠进行强迫游泳制模,模型成功后给予不同干预;干预24h后行为检测;之后快速处死,取脑固定,空白对照组不予任何处理,采用免疫组织化学方法检测大鼠大脑皮质NE和5-HT的表达。结果(1)NPY干预组和生理盐水干预组大脑皮质5-HT和NE的表达没有显著性差异(P>0.05),但二者均显著低于空白对照组表达(P<0.01);(2)NPY在干预后24h显示出抗抑郁样作用即大鼠强迫游泳不动时间开始缩短。结论NPY的抗抑郁样作用可能是通过调节大脑皮质NE和5-HT的表达而起作用的。

Abstract:
Objective To investigate effects of neuropeptide Y(NPY)on the expression of 5-hydroxytryptamine(5-HT)and noradrenalin(NE)in brain cordex by forced swimming rats in order to illuminate pathogenesis of depression.Methods Thirty male Sprague-Dawley rats were randomly divided into three groups:NPY administrated group (A),0.9% saline administrated group (B),and blank control group(C).After the development of the model of depression,the rats of group A and B were administered separately.At twenty-four hours after the injection,their behavior was recorded respectively.Then all the rats were sacrificed.Expression of 5-hydroxytryptamine and noradrenalin were observed by immunohistochemical assay.Results (1)The expression of 5-hydroxytryptamine and noradrenalin in cerebra cortex of group A and B was lower than that of group C.(2)The NPY administrated group showed significantly less immobility during the forced swimming test(P<0.01).Conclusion NPY may play a role in the process of anti-depression by regulating the expression of Noradrenalin and 5-hydroxytryptamine in cerebral cortex.
