摘    要:目的:建立高效液相色谱法测定血浆中佐匹克隆浓度。方法:采用Eurospher 100—5 C18色谱柱(200mm×4.6mm,5μm);流动相:0.02mol·L^-1磷酸二氢钾(含0.2%三乙胺,pH=4.0)-甲醇(38:62);流速:1.0ml·min^-1;柱温:3092;激发波长:315nm,发射波长:385nm。结果:血浆佐匹克隆在2.0~200.0ng·ml^-1浓度范围内线性关系良好(r=0.9995)。低、中、高3种浓度绝对回收率分别为92.64%,92.97%,94.66%,日内、日间RSD均小于10%。结论:本法可用于佐匹克隆血浆中浓度的测定以及药物动力学研究。

Determination the Concentration of Zopiclone in Human Plasma by HPLC

Abstract:Objective: To establish an HPLC method for the determination of zopiclone in plasma. Method: The Eurospher 100-5 C18 column(200mm × 4.6mm,5 μm)was used. The mobile phase consisted of 0.02mol·L^-1 potassium dihydrogen phosphate-methanol (38: 62). The flow rate was 1.0 ml·min^-1. The column temperature was 30℃. The excitation and emission wavelengths were 315nm and 385 nm, respectively. Result: The linear range of zopiclone was 2.0 -200.0ng·ml^-1( r = 0.999 5). The recoveries were 92.64%, 92.97% ,94.66% in the low ,meddle and high concentrations,respectively and RSD were below 10%. Condusion: The method can be used for the determination of zopiclone in plasma.
