摘    要:目的 探讨秦皮乙素对人肺癌H460细胞体外增殖与凋亡的影响.方法 将体外培养人肺癌H460细胞分为4组:空白对照组、秦皮乙素0.2 mg/ml组、秦皮乙素0.4 mg/ml组、秦皮乙素0.8 mg/ml组.给药干预48 h后,MTT法观察细胞增殖抑制率;流式细胞仪分析细胞凋亡率和周期;比色法检测Caspase-3的活性.结果 不同浓度的秦皮乙素均能抑制人肺癌细胞H460的体外增殖,并使细胞阻滞于S期,Caspase-3的活性上升,上述结果均呈剂量依赖性.结论 秦皮乙素能抑制人肺癌H460细胞的体外增殖,并能诱导H460细胞的凋亡,具有良好的抗肿瘤作用.

Abstract:Objective To investigate the effects of aesculetin on the proliferation and apoptosis of human lung cancer H460 cells in vitro. Method The human lung cancer H460 cell was cultured in vitro. Then the sample was divided into 4 groups as control group, aesculetin 0.2 mg/ml group, aesculetin 0.4 mg/ml group and aesculetin 0.8 mg/ml group, respec-tively. In 48 h after intervention, the cell inhibitory rate was observed by MTT assay, the cell apoptosis rate and cell cycle was analyzed by flow cytometry (FCM). The activity of Caspase-3 was detected by the colorimetry. Result Aesculetin could inhibit the proliferation of human lung cancer H460 cells in vitro significantly at all concentrations, cells were ar-rested in S phase, increasing the activity of Caspase-3, and all the effects observed were dose-dependent. Conclusion Aesculetin can inhibit the proliferation of H460 cells in vitro, and induce the apoptosis of H460 cells, suggesting a fair an-ti-tumor effect.
