FEZ1基因在食管鳞癌中的表达及意义

目的探讨肿瘤抑制基因FEZ1在食管鳞癌组织中的表达及其与食管鳞癌临床病理特征之间的关系。方法采用原位杂交法检测50例食管鳞癌及正常食管组织中FEZ1 mRNA的表达情况,采用Western blot法检测FEZ1蛋白在低分化食管癌细胞株EC-1和高分化细胞株EC9706中的表达,分析其与食管鳞癌患者各临床病理指标的关系。结果FEZ1 mRNA在食管鳞癌组织中的阳性表达率为42.0%,显著低于正常食管组织(86.0%)(P=0.000);随肿瘤分化程度的升高,FEZ1 mRNA的阳性表达率也逐渐增加(P=0.007),并且其在食管鳞癌组织中的阳性表达率与淋巴结转移密切相关(P=0.013)。Western blot结果显示FEZ1蛋白在食管癌低分化细胞株中的表达水平低于食管癌高分化细胞株,但差异无统计学意义(P=0.062)。结论在食管鳞癌组织中存在FEZ1基因的失表达,其缺失与食管鳞癌的分化和转移有关。

Significance of expression of FEZ1 gene in esophagealsquamous cell carcinoma

Objective To investigate expression of tumor suppressor gene FEZ1 and relationship between FEZ1 expression and clinicopathological features of esophageal squamous cell carcinoma.Methods The expression of FEZ1 mRNA in 50 cases of esophageal carcinoma and 50 cases of normal esophageal mucoma were detected by in situ hybridization(ISH).The expression of protein in esophageal carcinoma cell lines,EC-1 with low differentiation and EC9706 with high differentiation,was detected by Western blot.Results The positive expression rate of FEZ1 mRNA in esophageal squamous cell carcinoma(42.0%) were significantly lower than in normal esophageal mucoma(86.0%)(P=0.000).The positivity of FEZ1 mRNA in esophageal squamous cell carcinoma was related with differentiation(P=0.007) and lymph node metastasis(P=0.013).The expression of protein in low differentiated cell was lower than that in high differentiated cell,but there was no significant difference between them(P=0.062).Conclusion Missing expression of FEZ1 gene may be relation with differentiation and metastasis in esophageal squamous cell carcinoma.
