Abstract:AIM To study the effects of Platycodon grandiflorum polysaccharides on in situ intestinal absorption of platycodin D.METHODS Rats were randomly assigned into control group and low?dose,medium?dose,high?dose P.grandiflorum polysaccharides groups,the in situ intestinal perfusion model was established,after which the effects of P.grandiflorum polysaccharides on absorption rate constant(Ka),apparent absorption coefficient(Papp)and absorption fraction of oral medicine(fa)of platycodin D were investigated.After the perfusion,the rat intestinal segment was collected,and Claudin?1,Occludin and ZO?1 expressions were detected by immunofluorescence staining method.RESULTS Compared with the control group,the P.grandiflorum polysaccharides groups demonstrated increased Ka,Papp and fa values(P<0.05,P<0.01),and down?regulated Claudin?1,Occludin and ZO?1 expressions(P<0.05,P<0.01)in a dose?dependent manner.CONCLUSION P.grandiflorum polysaccharides can improve the in situ intestinal absorption of platycodin D,whose mechanism may contribute to the down?regulation of tight junction protein expressions.
