火焰原子吸收法测定志愿者单剂口服复方法莫替丁咀嚼片血清中钙镁浓度

摘    要:
目的采用火焰原子吸收法测定复方法莫替丁咀嚼片口服后血清中钙镁浓度,考察其体内过程以及对法莫替丁体内药代动力学过程的影响。方法健康志愿者10名,男女各半,于试验当日早7∶00空腹一次口服复方法莫替丁咀嚼片2片,于药前和药后取经时肘静脉血4ml,采用火焰原子吸收法测定血清中钙镁浓度。结果10名志愿者服药前后钙离子血浓度分别为7.138±0.0.436mg.L-1和7.530±0.331mg.L-1;镁离子血浓度分别为0.611±0.054mg.L-1和0.705±0.071mg.L-1。服药前、后,以及服药后各时间点钙镁离子血浓度方差分析显示,无统计学差异(P值分别为0.891和0.647)。结论提示复方法莫替丁咀嚼片中钙镁离子不经胃肠道吸收。

Abstract:
OBJECTIVE To determine the calcium and magnesium concentrations in human serum by flame atomic absorption method after a single oral dose of compound famotidine chewable tablets and to investigate their physiological disposition and whether influence the pharmacokinetic properties of famotidine.METHODS A single(two tablets) dose of compound famotidine chewable tablets were orally given to 10 healthy volunteers,half male and half female,at 7:00 am.4ml of blood was collected before and after administration at scheduled time.The concentrations of calcium and magnesium in serum were determined by flame atomic absorption method.RESULTS The serum concentrations of calcium before and after administration were 7.138±0.436mg·L~(-1) and 7.530±0.331mg·L~(-1),respectively.The serum concentration of magnesium were 0.611 0.054mg·L~(-1) and 0.705~0.071mg·L~(-1),respectively.The serum concentrations of calcium and magnesium before and after administration,and among different schedule timepoints after administration were no statistic difference by variance analysis(P was 0.891 and 0.647,respectively).CONCLUSION It was indicated that the calcium and magnesium contained in compound famotidine chewable tablets were not absorbed through gastrointestinal tract after oral administration.
