摘    要:
目的探讨改良荧光原位杂交(fluorescence hybridization,FISH)技术检测乳腺癌HER2基因的影响因素。方法回顾性分析总结65例改良FISH检测乳腺癌HER2基因中11例失败的原因。结果检测失败的11例中9例与酶消化有关,2例可能与组织固定有关。结论改良FISH检测HER2基因时要做好质量控制,蛋白酶K的消化时间控制最为关键。

Abstract:
Objective To explore the influential factors of improved fluorescence in situ hybridization (FISH) technique in the detection of HER2 gene in breast cancer tissues.Methods Materials were retrospectively analysed and summarized in 11 failure samples in 65 samples with breast cancer by improved FISH technique to detect HER2 gene.Results Nine failure detection was related to the proteinase K digestion time,2 failure detection may relate to tissue fixation.Conclusion Quality control should be strengthened to detect HER2 gene in breast cancer tissues by improved FISH technique,and the control on proteinase K digestion time is the most critical.
