摘    要:
目的探讨胰岛素对人乳腺癌细胞株MDA-MB-543生长的增殖作用。方法将胰岛素直接作用于体外培养的人乳腺癌细胞株(MDA-MB-543),采用四甲基偶氮唑盐(MTT)法检测细胞增殖,流式细胞仪测定细胞周期分布。结果胰岛素在1~10 mU/mL浓度范围内和作用时间2~16 h范围内,对MDA-MB-543增殖作用逐步增强,反映增殖活性的S期和G2/M期细胞比例逐渐增多,其中S期细胞比例与对照组比较有显著性差异(均P<0.05或0.01)。结论胰岛素能加快诱导MDA-MB-543细胞进入细胞周期S期增殖,呈剂量和时间依赖关系,但是短暂和可逆的。

Abstract:
Purpose To investigate effect of Insulin on the proliferation of human breast cancer cell line MDA-MB-543 growth.Methods The Insulin was directly used on human breast cancer cell line(MDA-MB-543) cultured in vitro,using MTT methods to detect cell proliferation and the distribution of cell cycle was determined by flow cytomery.Results In the 1-10 mU/mL Insulin concentrations and Insulin action time within the range of 2-16 h,the proliferation effect on MDA-MB-543 was gradually increased,and the proportion of S phase and G2/M phase cells which reflecting the proliferation activity was gradually increased,the proportion of S phase cells compared with the control group had significant differences(all P<0.05 or 0.01).Conclusion Insulin can speed up the induction of MDA-MB-543 cells getting into S phase cell cycle to proliferation,and is time-dependeut and dose-dependent,but it is transient and reversible.
