开心胶囊对犬心跳骤停复苏后心肌损伤的抑制作用

摘    要:【目的】观察开心胶囊对犬心跳骤停复苏后心肌损伤的作用。【方法】健康杂种犬15条,随机分为假手术组、模型组、开心胶囊组;复制心脏骤停及复苏模型,分别在恢复自主循环后5min、30min、90min、180min由颈内静脉插管采取中心静脉血,观测各时段肌钙蛋白T(cTnT)的含量变化;心肌组织切片在光镜和电镜下观察心肌损伤的结构变化。【结果】模型组犬血清cTnT在自主循环恢复后立即升高,各时段cTnT浓度与假手术组比较均有显著性差异(P<0.05);开心胶囊组在复苏后90min和180min时段cTnT浓度值均低于模型组(P<0.05)。光镜下模型组心肌染色阳性点总面积增大,与假手术组比较有显著性差异(P<0.05);开心胶囊组阳性总面积降低,与模型组比较有显著性差异(P<0.05);电镜下开心胶囊组虽然出现细胞结构损伤,但损伤程度较模型组轻。【结论】开心胶囊对犬心跳骤停复苏后心肌损伤具有良好的防治作用。

Abstract:Objective] To observe the effect of Kaixin Capsule (KC) on myocardial injury of dogs after resuscitation of cardiac arrest. Methods] Fifteen healthy hybrid dogs were randomized into three groups: pseudo-operation group (A), model group (B) and KC group (C). The model of resuscitation of cardiac arrest was duplicated. Five minutes, 30 min, 90 min and 180 min after recovery of autonomic circulation, central venous blood was taken from internal jugular vein by intubation method to observe the changes of troponin T (cTnT) content. Histological features of injured myocardium were examined under light microscope and electron microscope 180 min after recovery of autonomic circulation. Results] Serum cTnT content of group B increased promptly after recovery of autonomic circulation and differed from that of group A in different time (P<0.05). Serum cTnT content of group C was lower than that of group B 30 min, 90 min, 180 min after resuscitation (P<0.05). Positive-staining myocardial area was decreased and cellular impairment under electron microscope was mild in group C as compared with that in group B (P<0.05). Conclusion] KC exerts a better effect in preventing and treating myocardial injury after resuscitation of cardiac arrest.
