中药益坤宁对围绝经期大鼠卵巢雌激素受体表达的影响

摘    要:
目的观察中药益坤宁对围绝经期大鼠卵巢雌激素受体(estrogen receptor,ER)α、ERβmRNA和蛋白表达的影响。方法12月龄雌性Wistar大鼠随机分为老年模型组、西药利维爱组、中药益坤宁组,连续灌胃给药4周;以5月龄大鼠作为青年对照组。采用RT-PCR和免疫组织化学检测大鼠卵巢中ERα、ERβmRNA和蛋白的表达。结果RT-PCR和免疫组织化学结果都显示:老年模型组大鼠卵巢ERα、ERβmRNA和蛋白表达明显低于青年组(P<0.05),中药益坤宁组和西药利维爱组ERα、ERβmRNA和蛋白表达明显升高,与老年模型组相比有显著性差异(P<0.05或P<0.01)。结论中药益坤宁上调大鼠卵巢ERα、ERβ的表达,可能是治疗围绝经期综合征的作用机制之一。

Abstract:
Objective To study the effect of Yikunning on the expression of estrogen receptor α,β in rat ovaries during perimenopause period to investigate the mechanism of Chinese medicine Yikunning on perimenopausal syndrome. Methods 30 female Wistar rats of 12 month old were randomly divided into three groups: old control group, livial group, Yikunning group.10 female rats of 5 month old were selected as young control group, and gavaged with corresponding drugs for 4 weeks. The expression of ER α, β mRNA and protein were detected by RT-PCR and immunohistochemistry. Results The results of RT-PCR and immunohistochemistry showed that the expressions of ER α and ER β in ovaries were decreased significantly in aging model group, compared with that in young control group (P<0.01). The expressions of ER α and ER β in ovaries were significantly increased in Yikunning group and Livial group, compared with that in aging model group (P<0.05 or P<0.01). Conclusions Yikunning could up-regulate the expression of ERα and ER β, which may be one of the treatment mechanisms of Yikunning on perimenopausal syndrome.
