益气活血中药对放射性肺损伤大鼠血清及肺组织ET-1表达影响的实验研究

目的探讨血浆及肺组织中ET-1在放射性肺损伤中的改变及益气活血中药对其治疗作用。方法96只Wistar种雌性大鼠随机分为照射加中药组(A组)30只、照射加氨溴索组(B组)30只、单纯照射组(C组)30只、正常对照组(D组)6只。用直线加速器对前三组大鼠右肺进行分次照射(5Gy/次,1次/周,累积剂量为30Gy),于照射开始后第4、6、8周3个时间点分别处死三组大鼠各6只,采血测定ET-1浓度,肺组织测定ET-1免疫组化。结果照射后肺组织炎症和肺水肿进行性加重;C组照射后5,15,30,60d血清及肺组织ET-1水平进行性升高,A组、B组各时相值表达相似,均明显低于C组,差异具有统计学意义(P<0.05)。结论血浆ET-1水平的变化能反映放射性肺损伤的严重程度,益气活血中药能抑制肺组织ET-1的表达,降低血浆ET-1水平,可能是益气活血中药治疗放射性肺损伤的一个作用机制。

Experimental Study of Effect of Yiqi Huoxue Recipe on the Serum and Pulmonary ET-1 Expression in the Iung Injury Induced by Thoracic Radiation of Rats

Objective To explore the changes of endothelin-1 in plasma and lung tissues,and the curative effect of Yiqi Huoxue Recipe(Chinese traditional medicine of nourishing qi and activating blood)in radiation-induced lung injury.Methods96 wistar rats were randomly divided into irradiation group,irradiation with Yiqi Huoxue Recipe-treated group,irradiation with ambroxol-treated group and normal control group.The first three groups were irradiated at right hemithorax by linear accelerator with a dose of 30 Gy 6 Fraction 5 weeks.Animals were followed up for 26 weeks after the first irradiation.The contents of ET-1 in plasma and lung tissues were measured by ELISA and immunoreactivity repectively at the end of the 4th,6th,8th,12th,and 26th week after the first irradiation.ResultsPulmonary edema and pneumonia aggravated gradually in C group,pulmonary edema and pneumonia of A and B group were lighter than C group,the ET-1 levels in plasma and lung tissues increased rapidly in C group,the ET-1 levels in plasma and lung tissues of A group were similar to that of B group(P>0.05),and were both lower than C group(P<0.05).ConclusionET-1 could reflect the degree of radiation lung injury.Yiqi Huoxue Recipe could degrade the levels of ET-1 in plasma and lung tissues significantly.This might be one of their mechanisms in treatment of radiation-induced lung injury.
