摘    要:
 目的建立同时对小半夏汤中的5种核苷类成分进行含量测定的方法。方法采用Lichrospher C18柱(4.6 mm×250mm,5μm),以乙腈-水(1∶99)为流动相,体积流量1.0 mL·min-1,λ220~300 nm,柱温30℃。结果小半夏汤中的尿嘧啶、黄嘌呤、尿嘧啶核苷、次黄嘌呤核苷、鸟嘌呤核苷等的线性范围和相关系数分别为6.88~110.08 mg·L-1(r=0.999 6),2.82~45.04 mg·L-1(r=0.999 8),22.40~358.48 mg·L-1(r=0.999 9),1.96~31.32 mg·L-1(r=0.999 9),16.54~264.72 mg·L-1(r=0.999 8);加样回收率分别为100.31%(RSD=1.36%),100.21%(RSD=1.22%),99.98%(RSD=1.30%),100.44%(RSD=1.62%)和100.27%(RSD=1.36%);方法精密度、重复性良好,RSD均小于2.0%。结论本法操作简便、结果可靠、重现性好、检测快速、定量准确,可作为小半夏汤中核苷类成分的含量测定方法。

Abstract:
OBJECTIVE To establish a HPLC method for the simultaneous determination of five nucleosides in Xiao Banxia Tang.METHODS The samples were separated on a C18 column,and detected by a photodiode-array-detector.The mobile phase was acetonitrile-water(1∶99) at the flow rate of 1.0 mL·min-1.The detection wavelength was 220~300 nm,the column temperature of 30 ℃.RESULTS The linear ranges of uracil,xanthine,uridine,inosine,guanosine was 6.88-110.08 mg·L-1(r=0.999 6),2.82-45.04 mg·L-1(r=0.999 8),22.40-358.48 mg·L-1(r=0.999 9),1.96-31.32 mg·L-1(r=0.999 9)and 16.54-264.72 mg·L-1(r=0.999 8) respectively.The average recoveries were 100.31%,100.21%,99.98%,100.44% and 100.27% with corresponding RSDs of 1.60%,1.22%,1.30%,1.62% and 1.36%,respectively.The RSDs of precision and accuracy of the assay were not more than 2.0%(n=6).CONCLUSION The method is rapid,accurate,reliable and can be used to determine the nucleoside compounds in Xiao Banxia Tang.
