摘    要:目的探讨三七总皂苷(PNS)诱导大鼠骨髓间充质干细胞(MSCs)体外分化为心肌样细胞。方法采用骨髓细胞体外培养技术分离大鼠MSCs,用流式细胞仪检测细胞表面标志,用第8代MSCs进行体外心肌细胞诱导分化,采用相差显微镜、免疫组化及RT-PCR鉴定心肌细胞。结果大鼠MSCs细胞表面抗原CD44阳性,CD34阴性;经PNS体外诱导后细胞形态发生变化,其细胞免疫组化呈结蛋白(Desmin)、肌钙蛋白(cTnI)阳性;RT-PCR显示诱导后细胞转录肌球蛋白重链mRNA水平增加。结论大鼠MSCs体外在PNS诱导下可分化为心肌样细胞,为应用MSCs治疗心肌损伤提供了动物实验方法。

Abstract:Objective To explore the effect of Panax notoginseng saponins on the differentiation of rat bone marrow mesenchymal stem cells(MSCs) into cardiomyocyte-like cells in vitro. Methods Rat MSCs were isolated from adult bone marrow cells by culture in vitro, and their surface marks were detected by flow cytometry. MSCs of the 8th generation were used for the induction of differentiation of cardiomyocyte in vitro. Phase-contrast microscope,immunohistochemistry and RT-PCR were used for the identification of cardiomyocytes. Results The surface antigenic features of MSCs were positive for CD44 and negative for CD34.After induced by Panax notoginseng saponins, the morphology of MSCs changed,immunohistochemistry staining showed positive for desmin and troponin, and RT-PCR showed that the mRNA of myosin heavy chain (MHC) was highly expressed.Conclusion The rat MSCs can be differentiated into cardiomyocyte-like cells in vitro by Panax notoginseng Saponins induction. It provides an animal experimental evidence for using MSCs to treat myocardial damage.
