Losartan对血管平滑肌细胞增殖和血小板衍化生长因子及c-myc基因表达的影响

摘    要:
目的 探讨血管紧张素受体拮抗剂 Losartan 治疗高血压、动脉粥样硬化等心血管疾病的细胞作用机制。方法 运用免疫荧光组织化学技术和激光共聚焦显微镜定量观察 Losartan 作用下的血管平滑肌细胞( V S M C) 中细胞核增殖抗原( P C N A) 、 P D G F 和cmyc 原癌基因表达的平均荧光值。定量研究 Losartan 对体外培养血管平滑肌细胞增殖及血小板生长因子( P D G F) 和cmyc 癌基因表达变化的影响。结果  Losartan 作用下, V S M C 中的 P C N A、和cmyc 癌基因的平均荧光值均低于对照组,而 P D G F 的平均荧光值与对照组相比无显著差异。结论  Losartan 抑制 V S M C 的增殖及cmyc原癌基因的表达。该作用与癌基因调控的蛋白产物作用机制有关,是 Losartan 治疗高血压,参与血管重塑的重要的机制

Abstract:
Objective To investigate the mechanism of Losartan in treating hypertension and atherosclerosis.Methods The immunofluorescent quantitative skill and laser scanning confocal microscope (ACAS570) were employed to detect changes of the expression of PCNA, PDGF and c myc in cultured vascular smooth muscle cell effected through the Losartan .Results Under the observation of laser scanning confocal microscope (ACAS570), the fluorescent means of PCNA and c myc expression of VSMC in Losatan group were decreased compared with the control group, but that of PDGF had no changes.Conclusion Losartan possesses inhibitive effects on the proliferation of VSMC and the expression of c myc oncogene.This effect deals with the molecular mechanism of oncogene regulation.
