摘    要:目的在膀胱癌细胞系中过表达mir-200b,观察其对T24细胞迁移能力的影响,并检测WASF-3表达水平的改变。方法将mir-200b mimic,WASF-3 siRNA及阴性对照分别转染T24细胞,通过qPCR及Western印迹实验检测WASF-3表达水平的改变;利用Transwell实验检测转染后T24细胞迁移能力的改变。结果转染mir-200b及WASF-3 siRNA后WASF-3在mRNA及蛋白水平上均有明显下降,同时细胞迁移能力亦有明显下降。结论mir-200b能够通过改变WASF-3的表达水平抑制膀胱癌细胞系T24的迁移能力,为进一步探索miRNA在膀胱癌发生发展过程中的作用提供了研究基础。

Abstract:Objective To investigate the function of mir-200b in bladder cancer cell T24 migration and to detect the expression levels of WASF-3 in mir-200b transfected cells.Methods The mir-200b mimic,siRNA against WASF-3 or the negative control RNA duplex were transfected into T24 cells respectively.Then qRT-PCR and Western blotting were used to detect the expression of WASF-3.Transwell assay was used to detect the migration of T24 cells.Results Both of the mRNA and protein levels of WASF-3 were reduced in cells transfected with the mir-200b mimic and siRNA compared with the negative control.The migration was also inhibited in these cells.Conclusion mir-200b could inbibit the migration of bladder cancer cell T24 by reducing the expression of WASF-3.Further research is needed to explore the function of miRNA in bladder cancer generation and development.
