间接荧光抗体试验诊断实验性旋毛虫病的研究

摘    要:
本文应用间接荧光抗体试验比较了4种方法制备的旋毛虫幼虫抗原诊断实验性旋毛虫病的效果。结果表明,旋毛虫纯净幼虫冰冻切片抗原优于完整幼虫抗原、超声粉碎幼虫抗原及膈肌幼虫冰冻切片抗原。用此抗原检测感染旋毛虫的大鼠血清时,抗体阳性率为100%(13/13),而10份正常大鼠血清均为阴性反应。小鼠感染旋毛虫后2周即可检测到抗旋毛虫抗体,阳性率为11.11%(1/9),感染后5周抗体阳性率已升到100%(8/8),升高的抗体滴度可一直持续至感染后第15周。此抗原在-20℃至少可保存2.5年而不丧失其活性和特异性。应用滤纸血也可取得满意的检测结果。

Abstract:
he efficacy of Trichinella spiralis larva antigens prepared by 4 methods forimmunodiagnos is of experimental trichinos is was compared by indirect fluorescent antibodytest(IFAT).The results showed that the frozen section of the purified trichina larvae was su-perior to the whole larvae fixed on the slide with acetone,fragments of the larvae sonicatedand the frozen sections of the diaphragm muscle containing trichina larvae.When sera fromrats infected with Trichinella spiralis were tested by IFAT using the frozen sections of puri-fied larvae as antigen the antibody positive rate was 100%(13/13),sera collected from 10normal rats were negative reaction.Results with mice experimentally infected with Trichinel-la spiralis showed that the antitrichina antibodies could be detected 2 weeks after infectionwith the positive rate of 11.11%(1/9),increased to 100%(8/8) 5 weeks after infection.Ahigh antibody titer was sustained untill 15 weeks after infection. The antigen could be storedat-20℃ for at least 2.5 years without loss of its activity and specifity.The use of blooddried on absorbent paper in the test also gave excellent results.
