有氧运动训练对糖尿病大鼠Leptin-下丘脑NPY轴的影响

摘    要:本研究通过建立糖尿病大鼠有氧运动模型 ,探讨有氧运动训练对糖尿病大鼠瘦素 (Lep tin) -下丘脑神经肽Y(NPY)轴的影响。实验选用Wistar雄性成年大鼠 ,体重 195± 10g ,随机分为正常对照组、糖尿病对照组、糖尿病运动组。运动组每天游泳 6 0分钟 ,每周游泳 6天 ,连续游泳训练 6周。记录实验开始及结束时动物体重。测定血糖、血胰岛素浓度、内脏附睾脂肪垫重量、血清Lep tin及下丘脑NPY蛋白浓度和下丘脑弓状核NPY基因表达的非同位素原位分子杂交结果。结果表明 :长期有氧运动使糖尿病大鼠糖代谢紊乱得到改善 ,糖尿病大鼠食欲亢进减弱 ,摄食量减少 ,而内脏脂肪储量上升 ,体重增加。提示Leptin -下丘脑NPY轴可能起着重要的能量代谢调节作用

Abstract:The purpose of the present study is to investigate the effect of aerobic exercise on leptin-hypothalamus NPY in diabetic rats. The adult male Wistar rats (195±10g body wt) were adopted in this experiment. The rats were randomly divided into the following groups: normal control group; diabetic non-exercise group; diabetic exercise group. The food intake and body weights of the animals pre and post experiment were recorded. The rats were trained with swimming in exercise group,60min/day and 6 days/wk for 6 weeks. Serum insulin, blood glucose, the weight of epididymal fat pads (PED), serum leptin and the concentration of hypothalamus NPY were measured. The expression of NPY gene in arcuate nucleus (ARC) were determined by nonradioactive in situ hybridization. It was demonstrated that long-term aerobic exercise could improve glyco metabolism disorder and energy metabolism abnormality, abate excessive appetite, decrease the food intake, and increase splanchnic lipopexia and body weight in the diabetic rats, in which leptin- hypothalamus NPY may play an important part in the energy metabolism regulation.
