微粒包裹型幽门螺杆菌疫苗口服诱导小鼠粘膜免疫应答的研究

摘    要:
幽门螺杆菌(Helocobacter pylori,Hp)是慢性胃炎、消化性溃疡的主要病原菌,为分析Hp超声上清经微粒包裹后对小鼠的口服免疫效果,采用ELISA法检测血清、唾液、肠粘液的抗体改变情况,ELISPOT法分析派伊尔氏结(PP结)抗原特异性抗体形成细胞(ASC)的数量增减。发现微粒包裹型Hp疫苗免疫后可诱导较高的唾液sIgA水平和肠道sIgA反应,PP结抗原特异性抗体形成细胞(ASC)数量与肠道sIgA水平密切相关。

Abstract:
Helicobacter pylori is a major cause of chronic antral gasritis and peptic ulcer diseases.To analysis the mucosal immune responses in BALB/c Mice by oral immunization with H.pylori lysates encapsulated in copolymers of poly(D,L-lactide) and polyethylene glycol(PLEA) microparticles,we assayed antibody production in salivary and gut washing by ELISA and antibody secreting cells in Peyer's Patches(PP) by ELISPOT.Oral immunizations induced more significant H.pylori-specific intestinal IgA response as well as salivary IgA response than those detected with unencapsulated soluble antigen(P<0.05).The presence of antibody-secreting cell in PP was correlated with IgA level in gut washing .These data suggested that PLEA-Hp could induce H.pylori-specific mucosal immune responses in vivo and locally synthesized specific sIgA antibodies.
