羟喜树碱纳米制剂对原位肝肿瘤小鼠药效学研究

摘    要:目的观察羟喜树碱纳米制剂对原位肝癌小鼠的抗肿瘤作用。方法造模小鼠30只随机分成3组,每组10只,羟喜树碱纳米组予羟喜树碱纳米制剂5 mg.kg-1;羟喜树碱粉针剂组予羟喜树碱冻干粉针剂5 mg.kg-1;对照组予等量生理盐水,每周d 1、4尾静脉给药,共2 wk。观察原位肝癌的生长状态及组织病理学改变、计算抑瘤率、MTT法检测细胞增殖抑制率及流式细胞仪检测细胞凋亡率。结果与对照组比较,羟喜树碱粉针剂组及纳米组均可抑制小鼠原位肝癌的生长,其羟喜树碱纳米组抑瘤率为48.76%,高于粉针剂组(38.51%);羟喜树碱纳米组对肝癌细胞及正常肝细胞的增殖抑制率分别为82.63%、27.10%,对瘤组织细胞凋亡率为(49.72±7.51)%。结论羟喜树碱纳米制剂可抑制小鼠原位肝癌生长。

Abstract:AIM To evaluate the efficacy of nano hydroxycamptothecin(HCPT) in the treatment of mouse liver cancer model.METHODS Thirty mice were randomized into three groups(n = 10 each group),which were respectively administered with nano HCPT,HCPT and sodium chloride(control group)5 mg·kg-1 for two weeks at the first and the fouth day of each week.The growth of implanted tumors and the pathological changes were observed.The rate of tumor inhibition was calculated according to tumor size.The tumor cell growth inhibition rate was measured by MTT assay,and the apoptosis rate was determined by flow cytometry.RESULTS Compared with the control group,HCPT group and nano HCPT group inhibited the growth of liver cancer in situ,and the inhibition rate of nano HCPT group(48.76%)was higher than that of the HCPT group(38.51%).The inhibition rate to tumor cells and to liver cells in nano HCPT group was 82.63% and 27.10%,and the apoptosis rate of nano HCPT group was(49.72 ± 7.51)%.CONCLUSION Nano hydroxycamptothecin is effective in the treatment of mouse liver cancer.
