| 摘 要： | [目的]研究Celecoxib对肺癌A549细胞生长和凋亡的影响并探讨其作用机制.[方法]用Western blot法检测Celecoxib抑制肺癌A549细胞后生存素（Survivin）表达的变化;用酶联免疫吸附法（ELISA）测定培养上清液中前列腺素2（prostaglandin-2,PGE2）含量的变化;用四唑盐（MTT）比色法检测细胞增殖;流式细胞仪测定细胞周期.[结果]随着Celecoxib剂量的增加和时间的延长,PGE2的含量减少,survivin表达也相应减少,细胞增殖减少而凋亡增加.[结论]Celecoxib可以通过PGE2的途径下调A549细胞survivin的表达并促进细胞凋亡. |

| Abstract: | [Purpose] To investigate the effect and mechanism of Celecoxib in inducing proliferation inhibition and apoptosis in human lung cancer A549 cells.[Methods] The expression of survivin through Celecoxib blockading were detected by Western Blot.ELISA were used to detect the change of prostaglandin-2(PGE2)content in the supernatant of culture solution.The anti-proliferative effect was measured by using Methabenzthiazuron(MTT)assay.Cell cycle and apoptosis were analyzed by using flow cytometry.[Results] With the blockade of Celecoxib increased,the PGE2 content reduced and the expression of survivin weakened,but the apoptosis rised.[Conclusion] Celecoxib can down-regulate the expression of survivin and then promote the apoptosis of A549 cells via PGE2 pathway. |
