神经肽Y对培养的自发性高血压大鼠动脉平滑肌细胞的影响

摘    要:
应用细胞培养,~3(H-)胸腺嘧啶核苷和~3(H-)亮氨酸的掺入及细胞计数方法,研究缩血管物质神经肽Y(NPY)对血管平滑肌细胞生长的作用。结果发现,用神经肽Y作用48h后可促进离体培养的自发性高血压大鼠(SHR)动脉平滑肌细胞DNA合成增加28.0%,蛋白质合成增加54.6%,与对照组比较有显著性差异(P<0.01);而细胞数无明显增加(P>0.05)。提示,NPY对培养的SHR动脉平滑肌细胞有促肥大效应;SHR血管壁上NPY分布密度增加是引起高血压时血管壁结构改变的重要因素之一。

Abstract:
We applied the method of cellular culture, tritiated thymidine and leucine incorporation, and calculation in cell number to investigate the role of vasoconstrictor substance (neuropeptide Y NPY) in vascu-lar smooth muscle cell (VSMC) growth. It was observed that 48-hour exposure to neuropeptide Y for these cells caused an 28. 0 % and 54. 6 % increase respectively in DNA and protein synthesis in cultured VSMC in SHR (compared with serum control P<0, 01), but no significant increase in cell number (P>0. 05). The re-sults demonstrate that NPY accelerates VSMC hypertrophy in SHR, and an increase in density of NPY-con-taining innervation in the vasculature of SHR is one of the important factors in the structural changes of hy-pertensive vessels.
