铁岭市一起皮肤炭疽疫情中炭疽芽胞杆菌的快速检测与分析

摘    要:
目的 对2015年7月铁岭市发生一起皮肤炭疽疫情中运用UPT上转发光免疫分析仪联合荧光定量PCR技术进行描述与分析,探讨可以快速现场检测炭疽芽胞杆菌的方法。方法 采集疑似皮肤炭疽患者的焦痂下涂抹样本10份和炭疽疫区外环境样本19份,运用上转发光免疫层析技术联合荧光定量PCR技术对标本进行检测,与此同时进行细菌分离培养。结果 10份患者的痂下涂抹物样本和2份病死牛血污染土壤样本在上转发光免疫层析技术和荧光定量PCR技术检测结果均呈现阳性。细菌分离培养技术仅分离到1株炭疽芽胞杆菌。结论 实验室检测结果充分证明此次疫情的病原体为炭疽芽胞杆菌的感染。上转发光免疫层析技术和荧光定量PCR方法的联合应用,能快速、特异、灵敏检测出炭疽芽胞杆菌,具有推广应用价值。

Abstract:
Objective In July 2015, a case of skin anthrax in Tieling was analyzed by UPT chemiluminescence immunoassay analyzer combined with real-time PCR technology, and a method for rapid detection of Bacillus anthracis was discussed. Methods 10 samples of cutaneous eschar smear and 19 samples of anthrax epidemic environment were collected from suspected anthrax patients, using up-converting phosphor technology combined with real-time PCR technology to detect the samples by bacterial culture at the same time. Results 10 patients under eschar smear samples and 2 dead cattle contaminated soil sample subjected to up-converting phosphor technology combined with real-time PCR technology demonstrated positive results. One strain of Bacillus anthracis was isolated from bacteria isolation and culture. Conclusion The laboratory test results fully demonstrated that the pathogen of the epidemic was infected by Bacillus anthracis. The combined application of up-converting phosphor technology and real-time PCR technology can detect Bacillus anthracis rapidly, specifically and sensitively, and has wide application value.
