恒河猴促滤泡激素生物测定法的建立和应用

摘    要:目的建立恒河猴促滤泡激素(FsH)生物测定法。方法利用FsH体外生物测定法,测定了恒河猴血清中FSH含量。结果测定方法的灵敏度为0.02ng/管.批内和批间误差分别为(2.53±1.25)%和(4.02±2.17)%,证明该方法准确性高;恒河猴血清中FsH含量反应曲线和hFSH标准曲线平行良好。说明该方法可以用来检测恒河猴血清中的具有生物活性的FsH变化.反映了动物正常的生理状态;回收实验表明.在该测定方法中加入5ng、10ng、20ng hFSH时的回收率分别为(101.28±5.66)%、(99.78±4.51)%、(100.61±5.42)%,说明测定结果的可靠性。结论本实验所建立的FSH体外生物测定法灵敏度高.稳定性强。

Abstract:Objective To establish a bioassay of follicle stimulating hormone (FSH) in serum of rhesus monkey. Methods The FSH content of the rhesus monkey was measured by bioassay in vitro. Results The sensitivity of the assay was 0.02 ng/per tube, the intra and inter assay variations were (2.53± 1.25)% and (4.02±2.17)%; both the dose-response curve of FSH in serum of rhesus monkey and the calibration curve of hFSH are in good linearity which reflected the pliysiological status of the animals; the recoverable rates of 5, 10 and 20 ng hFSH added to the diluted ram serum were (101.28±5.66)%, (99.78±4.51)% and (100.61 ±5.42)% respectively. Conclusion The bioactive FSH level in pituitary extract is by far higher than that in circulation.
