裸鼠皮下-肝原位移植人肝细胞肝癌模型的建立和抑癌基因PTEN的表达

摘    要:[目的]探索应用人类肝细胞肝癌细胞株建立裸鼠皮下-肝脏原位肝癌模型,及抑癌基因PTEN在该模型中的表达。[方法]应用Bel-7402肝癌细胞株建立裸小鼠皮下-肝脏原位肝癌模型48只,12只正常裸小鼠作对照;采用二步法免疫组化方法检测PTEN的表达。[结果]4周后解剖结果证实成模率100%;与正常组对比,模型组动物生存时间明显缩短,肝脏和脾脏明显增大(P<0.05或<0.01)。瘤体积与模型生存时间负相关,与脾指数正相关(P<0.01)。模型表现出明显的肝脏炎症反应和瘀胆。肝内肿瘤转移率80.0%,肺内肿瘤转移率10.4%。PTEN基因表达强度:癌旁组织>正常肝组织>癌组织(P<0.01)。[结论]该模型用于研究人类原发性肝癌,造模时间短、成瘤率高,生物学行为与人类肝癌接近,肿瘤体积过大是瘤鼠致死的重要因素。

Abstract:[Objective]To establish human hepatoma model from subcutane to orthotopic transplantion with Bel-7402 in athymic mice and observe PTEN expression.[Methods]Forty eight male athymic mice models were established with Bel-7402 from subcutane to orthotopic transplantion.And another twelve rats were as normal control.Expression of PTEN was detected by Envision's two step method.[Results] Four weeks later,HCC occurred in all of the 48 mice.Compared with control group,these models had shorter survival time,larger livers and spleens,P<0.05 or <0.01.There was negative correlation between volume of tumors and survival time and existed positive correlation between tumors volume and spleen exponent(spleen weight/body weight),P<0.01.Obvious inflammatory reaction and cholestasis in livers were observed in model group.The rate of tumor metastasis within livers of models was 80.0% and to lung was 10.4%.The result of immunohistochemistry indicated that the intensity of PTEN in latero-cancer tissue was higher than the hepatic tissue and cancer tissue,P<0.01.[Conclusion]It is a good method to establish human hepatoma model.The time is short and achievement ratio is high,further more,biology behavior is close to primary hepatic carcinoma.Large tumor volume is a main death cause of the mice in model group.
