摘 要: | 目的研究大鼠血管狭窄处内皮细胞组织因子(tissue factor,TF)的表达规律。方法将45只SD大鼠(重量320±18.5 g)随机分为对照组和颈动脉狭窄组,狭窄组又分为0.25、0.5、1、2、4、8、16、24 h,8个时相,套扎法建立左颈总动脉狭窄模型,应用数字减影血管造影和多普勒血流仪在体测定切应力及血流量。术后不同时相点用原位杂交和免疫组化法,检测TF的mRNA和蛋白表达。应用图像分析系统,测定内膜平均灰度,进行统计学分析。结果正常对照组内皮细胞TF的mRNA转录和蛋白合成微弱;狭窄15 min后,内皮细胞胞浆TF基因mRNA转录和蛋白合成升高,与对照组比较均有显著性差异(P0.01)。TF基因mRNA转录和蛋白合成,4 h达到峰值,与邻近组比较具有显著性差异(P0.01)。结论血管狭窄后,切应力能够早期诱导动脉狭窄处内皮细胞TF基因表达。

Abstract: | Objective To investigate the expression of tissue factor(TF) in endothelial cells induced by fluid shear stress in vivo.Methods 45 male SD rats(weight 320 ± 18.5 g) were randomly divided into control group and carotid artery stenosis,stenosis group was divided into 0.25,0.5,1,2,4,8,16,24 h,8 time phase,ligation of the left common carotid artery stenosis model,determination of shear stress and blood flow.After different time points by using in situ hybridization and immunohistochemistry to detect the expression of mRNA,TF and protein,with an image analysis system,determination of the average gray intima,statistical analysis.Results The mRNA and potein of TF was faintly expressed in intima of control group,after exposed to stenosis,the staining intensity was increased 15 minutes after wall shear stress,The mRNA and protein expresion of TF markedly increased comparing with control group (P 〈 0.01).The mRNA and potein of TF peaked at 4 hour,significant differences were present between vicinity group (P 〈 0.01).Conclusion Carotid artery stenosis,shear stress can express artery stenosis endothelial cell induced by TF gene in early. |
