氨茶碱、地塞米松对哮喘患者CD4+CD25+T调节细胞抑制功能的影响

摘    要:目的探讨哮喘患者CD4+ CD25+ T调节细胞的作用是否存在缺陷及药物(地塞米松、小剂量氨茶碱)对其作用的影响,为有效控制哮喘提供依据。方法分离哮喘患者及健康对照者外周血CD4+ CD25+ T调节细胞及CD4+CD25-T淋巴细胞并培养,设CD4+ CD25+ T调节细胞组、CD4+CD25-T淋巴细胞组及CD4+ CD25+ T调节细胞联合CD4+CD25-T淋巴细胞组,第3组又分为氨茶碱干预组、地塞米松干预组及空白对照组,72h后取培养上清液用ELISA法检测IFN-γ、IL-5、IL-13水平。结果健康对照者CD4+ CD25+ T调节细胞可抑制IFN-γ、IL-5及IL-13的生成(P<0.01),哮喘患者仅抑制IFN-γ、IL-5的生成(P<0.01);地塞米松预处理CD4+ CD25+ T调节细胞后,健康对照者CD4+ CD25+ T调节细胞抑制IFN-γ(P<0.05)、IL-5(P<0.01)、IL-13(P<0.01)生成能力增强,哮喘患者抑制IL-5、IL-13生成能力增强(P<0.01);采用氨茶碱预处理后,健康对照者CD4+ CD25+ T调节细胞抑制IL-5(P<0.01)、IL...
