高山红景天黄酮类化合物的分离与鉴定

摘    要:
目的对高山红景天药材的化学成分进行研究。方法采用反复硅胶柱色谱、ODS柱色谱、Sephadex LH-20柱色谱和半微量制备高效液相色谱分离纯化,根据ESI-MS、1H-NMR和13C-NMR等谱学数据进行结构鉴定。结果从高山红景天根和根茎的乙醇提取物中分离得到8个单体化合物,分别鉴定为黄酮类化合物herbacetin 3-O-[α-L-rhamnopyranosyl(1→2)-β-D-glucopyranosyl]-7-O-α-L-rhamnopyranoside(1)、三叶豆苷(trifolin,2)、草质素苷(rhodionin,3)、山奈酚(kaempferol,4)、山奈酚7-O-α-L-鼠李糖苷(kaempferol7-O-α-L-rhamnopyranoside,5)、小麦黄素(tricin,6)和大花红景天素(rhodiolinin,7);另外还得到2(3H)-苯并噻唑硫酮(2(3H)-benzothiazolethione,8)。结论化合物1为新化合物,化合物2和8为首次从景天科植物中分离得到。

Abstract:
Objective To study chemical constituents of root and rhizome of Rhodiola sachalinensis A. Bor. Methods The chemical constituents were isolated by silica, Sephadex LH-20, ODS column chromatography and HPLC. Chemical characters and spectroscopic analysis were employed for the structural identification. Results Eight compounds were obtained and their structures were identified as: herbacetin 3-O-[α-L-rhamnopyranosyl(1→2)-β-D-glucopyranosyl]-7-O-α-L-rhamnopyranoside (1), trifolin (2), rhodionin (3), kaempferol (4), kaempferol 7-O-α-L-rhamnopyranoside (5), tricin (6), rhodiolinin (7) and 2(3H)-benzothiazolethione (8). Conclusions Herbacetin 3-O- [α-L-rhamnopyranosyl (1→2)-β-D- glucopyranosyl]-7-O-α-L-rhamnopyranoside (1) is a new flavonol glucoside which has not been reported before. Compounds 2 and 8 are isolated from family Crassulaceae for the first time.
