补阳还五汤对代谢综合征大鼠海马IR、IRS-1mRNA表达的研究

摘    要:
目的:研究补阳还五汤对代谢综合征大鼠海马IR、IRS-1mRNA表达的影响。方法:以高脂高糖高盐饮食联合小剂量链脲佐菌素(STZ)建立代谢综合征大鼠模型,随机分为模型组、中药补阳还五汤组、西药罗格列酮组,另设正常对照组。给予相应受试药物,周期8 w。实验结束时,腹主动脉采血,取海马组织。检测各组血糖、血脂、双抗体夹心ELISA法测定血清CRP,实时荧光定量PCR技术检测海马IR、IRS-1mRNA的表达水平。结果:补阳还五汤组大鼠的血糖、血脂、血清CRP含量较模型组均显著降低,海马IR、IRS-1mRNA的表达水平较模型组均显著升高(P〈0.05)。结论:降低血糖、血脂,降低血清CRP,升高海马IR、IRS-1mRNA的表达,减轻中枢胰岛素抵抗可能是补阳还五汤干预代谢综合征认知功能障碍的部分作用机制。

Abstract:
Objective: To research the mechanisms of Bu-yang-huan-wu decoction that affect IR, IRS-lmRNA expression in hippocampus tissue of metabolie syndrome rats. Methods: Established metabolic syndrome rats model by high in fat and sugar, high salt diet in combination with small dose of chain urea with cephalosporins (STZ) . Departed metaboliesyndrome rats as model group, rosiglitazone group and Bu-yang-huan-wu decoction group act as the normal group. Treated for 8 weeks, detect the blood glucose, blood fat, blood secum C reactive protein (CRP), IR, IRS-lmRNA expression in hippocampus tissue by real-time PCR. Results: Bu-yang-huan-wu decoction can degraded the blood glucose, fat and blood CRP, elevate the IR, IRS-1 mRNA expression in hippo- campus tissue (P 〈 0.05) . Conclusion: Inhibit the blood glucose, fat and blood CRP, elevate the IR, IRS-lmRNA expression in hippocampus tissue, restrain central insulin resistance maybe one of the possible curing mechanisms that prevented metaboliesyndrome cognitive impairment by Bu-yang-huan-wu decoction.
